# Moderna Stock Before ESMO 2026: The Data Can Prove a Melanoma Drug, the Price Assumes a Platform — InvestMoat Research

_Moderna had more than tripled from its pre-readout close by October 16, and its October 24 ESMO data can show how good a melanoma drug intismeran is, but I think the price is paying for an mRNA cancer platform that only a randomized win in a second tumor type can prove._

## Summary

Moderna closed at $62.96 on August 18, 2026, the day before Merck and Moderna said intismeran plus KEYTRUDA met its Phase 3 endpoints in resected melanoma, and at $218.01 on October 16, a week before the full data at ESMO. I argue the October 24 data can settle the size of a melanoma product, while the October price already sat above a base case that assumes a supportive result in a second tumor type. In this healthcare cohort the framework rates regulatory lock-in strong only at Regeneron and Vertex, whose approvals rivals can't match; Moderna's respiratory approvals are of a kind its rivals hold, and they didn't stop revenue falling from $19.3 billion in 2022 to $1.9 billion in 2025. A randomized win outside melanoma is what would change that, and BioNTech's colorectal stop in August shows that result isn't guaranteed.

## What would prove this wrong

By December 31, 2027, a randomized intismeran study outside melanoma (the fully enrolled Phase 2 trials in adjuvant kidney or bladder cancer, or a Phase 3 in lung cancer) meets its primary endpoint. That would mean the platform was proven on roughly the timetable the October 2026 price assumed, and the discount I argue for a second tumor type was too steep.

MRNA price: (live) · Close before the Phase 3 release: $62.96 · Where the August notes convert: $210.58 · Full Phase 3 data: Oct 24

Moderna was the largest decliner in the S&P 500 on Friday, October 16. It closed down 7.8% at $218.01. That was still about three and a half times its August 18 close, the night before Merck and Moderna said their individualized cancer therapy, intismeran, had beaten KEYTRUDA alone in a Phase 3 melanoma trial. The full results are presented on October 24 in the Presidential Symposium at ESMO in Madrid, and I'm writing this before them on purpose, so the claims below can be checked against what comes out.

Most coverage treats October 24 as a binary event on the melanoma data. The sell side mostly sits at Hold, with a consensus target well below the October 16 close, so the disagreement I'm interested in isn't whether the stock is expensive. It's what the buyers who took it from $174 to $236 without new efficacy data think they own. I think they're paying for mRNA as a cancer platform, and I think October 24 can't tell us whether it is one.

The MRNA stock page sets out the scenario ladder, and the scorecard further down carries its numbers live. The base case there isn't melanoma alone. It already assumes a melanoma filing in 2027 plus a supportive result in kidney or bladder cancer. On October 16 the stock closed well above that base, closer to the bull case, which needs a second tumor type to hit a registrational bar. So even on the site's own ladder, the October price was paying for more than one cancer drug.

There's a fair reading of the run that's only about melanoma. ESMO doesn't put weak data in its Presidential Symposium, and the slot was announced on September 21, in the middle of the climb. Buyers may simply have read it as a sign of a large melanoma effect. I think that's partly right, and I come back to it in the counter-case. Even a large melanoma effect, though, makes the melanoma product bigger. It doesn't add a second product, and the price was already past the case that has one.

Every positive intismeran result is in melanoma, and every one pairs it with KEYTRUDA. Melanoma is one of the cancers where checkpoint drugs work best, which is part of why it was tested first. KEYNOTE-942's five-year numbers are strong and a Phase 3 win in more than a thousand patients is a real result, but both show the effect where it was most likely to show up. The precedent row matters too. KEYTRUDA and Opdivo were both approved in adjuvant melanoma on recurrence-free survival, so I'd treat a filing on the INTerpath-001 data as likely rather than as an upside surprise.

BioNTech's colorectal trial is the nearest thing to a test somewhere else, and it went the other way. It gave a rival individualized mRNA therapy on its own, without a checkpoint drug, in a cancer where checkpoint drugs mostly don't work, and it was stopped in August. BioNTech's pancreatic trial, which adds checkpoint inhibition and chemotherapy, continues. Moderna's stage I lung Phase 3 being presented at ESMO has an arm that gives intismeran alone against placebo, alongside the arm that pairs it with Merck's under-the-skin KEYTRUDA, so Moderna is running that same monotherapy experiment. I don't read BioNTech's result as evidence against intismeran. I read it as evidence that a second tumor type is a genuine experiment whose result depends on the tumor and the partner drug as well as the mRNA.

## Which drug moats the framework rates strong

To see what a proven platform looks like in the framework's terms, I put Moderna next to the four other drug makers in coverage and read the pillars that carry a drug company's protection: its approvals, its clinical data, its process know-how, how embedded its products are in treatment, and its scientists. I've grouped them by whether a rival holds the same kind of approval, because that turns out to be what the framework's notes hinge on.

**The pillars that protect a drug maker's revenue, grouped by whether a rival holds the same kind of approval. Statuses are read live from each stock's analysis.**

| Ticker | Regulatory Lock-In | Proprietary Data | Business Logic | Transaction Embedding | Talent Scarcity |

Regulatory lock-in is rated strong at Regeneron and Vertex and intact at the other three. The notes explain why, and it isn't the number of approvals. Lilly has many approved drugs across diabetes, obesity, oncology and Alzheimer's, and its GLP-1 approvals still rate intact because Novo holds the same kind. Novo's rate intact for the mirror reason, and its embedding in treatment is rated weakened because Lilly now wins most new prescriptions. Regeneron's strong rating rests on antibodies like Dupixent and Eylea HD that nobody else sells, and Vertex's rests on a cystic fibrosis franchise with no real competitor. Moderna's note puts it with Lilly and Novo on this pillar: its respiratory approvals are of a kind Pfizer and BioNTech, GSK and Sanofi also hold, and intismeran isn't approved yet.

Nothing gets approved on October 24, so the data can't move Moderna's regulatory rating. What they can do is make a melanoma approval more likely and tell us how big that product will be. On my reading of the notes, getting to strong would take an approval rivals can't match, and for an individualized cancer therapy the obvious candidate is a second tumor type where nobody else has one.

> **The test I'd apply** — When a biotech is priced as a platform, I'd look for approvals rivals can't match rather than the number of trials that worked. Regeneron and Vertex have several. Moderna's respiratory approvals are matched by its competitors, and its cancer therapy has one positive Phase 3 in one tumor type, so on that test the platform claim is still open.

## Why a second tumor matters more than a better hazard ratio

- The design process carries over to a new tumor, but the result may not. Each dose is made from one patient's tumor mutations, and the same sequencing, design and manufacturing run serves every cancer. Whether the immune response it triggers stops a recurrence depends on the tumor, and BioNTech's colorectal stop is a recent case where it didn't.
- The partner drug and half the profit are Merck's. Merck exercised its option in 2022 for $250 million, and the two companies share costs and profits equally worldwide. Regeneron shares Dupixent with Sanofi, so a partner doesn't rule out a strong rating, but Regeneron also has approved antibodies it keeps to itself.
- Manufacturing is one batch per patient. That's hard to copy, which is part of the moat, and it's also what has to scale before melanoma revenue shows up. A stronger hazard ratio raises demand for that capacity without adding any.
- Financing is off the table for now. The $2.6 billion of notes, on top of $6.9 billion of cash and investments at June 30 and less the $950 million settlement paid in July, mean Moderna doesn't need a good October 24 to fund the next readouts, so the price will be judged on the data.

## Moderna already has a platform with several approvals

The strongest objection is that I'm grading Moderna on cancer alone. Its mRNA platform has already produced several independent approvals: Spikevax, mNEXSPIKE, mRESVIA, and mFLUSIVA, which FDA approved on August 5, 2026 as the first mRNA flu vaccine. The same lipid delivery and manufacturing know-how carried across all of them. Someone holding this view would say the platform question was settled years ago and cancer is the next application.

**Moderna's approved US products and the rivals that sell vaccines against the same disease. Approvals as Moderna reported them; the rivals column is my summary of the main competing manufacturers.**

| mNEXSPIKE | COVID-19 | Approved; one of three approved products entering 2026 | Pfizer and BioNTech, Novavax |
| mRESVIA | RSV | Approved; one of three approved products entering 2026 | GSK, Pfizer |
| mFLUSIVA | Influenza | Approved Aug 5, 2026 for adults 50+, accelerated approval for 65+ | Sanofi, CSL Seqirus, GSK and others |

My answer is that the platform produced the approvals and the approvals didn't protect the revenue. Total revenue went from $19.3 billion in 2022 to $1.9 billion in 2025. Much of that is the pandemic ending, which no moat would have stopped. The rest is that Pfizer and BioNTech, GSK, Sanofi and others sell vaccines against the same diseases, so a Moderna approval competes on price and timing every season. That's the same thing that keeps Lilly's approvals intact in the matrix, and Lilly is the useful reminder here that a long list of approvals isn't what earns a strong rating.

The second version of the bull case is better, and I take it seriously. Cancer isn't flu. Nobody else has an approved individualized cancer therapy, and if intismeran works in melanoma the next tumor uses the same process and the same partner, so the odds in a second tumor are better than for an unrelated drug. Kidney and bladder are both cancers where KEYTRUDA already works after surgery, unlike colorectal, so BioNTech's failure may simply not apply. If the kidney Phase 2 reads out positive, the stock will have been early rather than wrong. That's why my falsifiable claim is about the second tumor and not about October 24.

The third is the Presidential Symposium reading. If the hazard ratio on October 24 is far better than KEYNOTE-942's and overall survival is already trending the right way, the melanoma product alone could be worth much more than the stock page's ladder assumes, and the October price might be a fair bet on melanoma rather than an early bet on a platform. I can't rule that out before seeing the data. It would mean I've got the size of the first product wrong, which is a different mistake from the one I'm warning about, and the scenario ladder on the stock page would need re-striking upward.

> **What would break this** — In order of how likely I think it is. The kidney or bladder Phase 2 reads out positive in 2027 and a lung Phase 3 follows, which would prove the platform on roughly the timetable the October price assumed. The October 24 data show a melanoma effect large enough that melanoma alone justifies the price, which would mean I misjudged the first product rather than the platform. Merck and Moderna restructure the deal so Moderna keeps more than half the profit, which would change what each approval is worth to Moderna's shareholders.

I'd use the same test on any drug platform the market gets excited about, and count the approvals rivals can't match rather than the trials that worked. Moderna's mRNA platform has several approvals in vaccines that rivals do match, and one positive Phase 3 in cancer. Until a second cancer result comes in, I'd treat whatever the stock trades at above a melanoma-only value as an option on the platform that the market bought on August 19.
